Abstract

The carbohydrate structure of a proline-rich glycoprotein from human parotid saliva is presented. Studies were performed on tritiated glycopeptides prepared by extensive pronase digestion of galactose oxidase/sodium borotritide treated glycoprotein. Structural characterization was carried out using a combination of periodate oxidation, methylation analysis, nitrous acid deamination, and glycosidase treatment. A triantennary asparagine-linked structure was found which contained peripheral Galβ1→4 (Fucα1→3) GlcNAc sequences.
