Abstract

Nonalcoholic fatty liver disease (NAFLD) and nonalcoholic steatohepatitis (NASH) are leading causes of liver diseases in children as well as adults. Oxidative stress is recognized as an underlying mechanism for the progression of this condition. Several studies are being conducted with an aim to explain molecular pathological mechanism of NASH that could help identify important targets for therapeutic or preventive interventions for NASH patients. Paraoxonase (PON) 1 is an enzyme that is known for its role as an antioxidant in cardiovascular disease. This enzyme is synthesized in liver and is found in circulation bound to HDL. Recent studies have investigated its role in liver diseases, such as chronic hepatitis, hepatocellular carcinoma, acute viral hepatitis, chronic alcoholic disease, and NASH. This chapter summarizes PON enzymes structure and function, and its role in liver diseases that are mediated through oxidative stress.
