Relationship between expression of specificity protein 1 of colorectal adenocarcinoma and clinicopathological parameters

Abstract

Objective: To investigate the expression changes of specificity protein 1 (Sp1) in colorectal adenocarcinoma tissues and its relationship with the clinicopathological parameters of patients.

Methods: The levels and differences of Sp1 mRNA of colorectal adenocarcinoma tissues and normal tissues were analyzed by retrieving the mRNA array database of Oncomine. The expression of Sp1 protein of 65 colorectal adenocarcinoma tissues and adjacent normal tissues was detected by the immunohistochemistry (IHC) assay. The expression of Sp1 protein of colorectal cancer cell lines of 5 different malignant degrees was detected by the Western blotting. The relationship of the expression level of Sp1 and the clinicopathological parameters of patients with colorectal adenocarcinoma was analyzed.

Results: There was no significant difference of Sp1 mRNA level between colorectal adenocarcinoma tissues and normal tissues according to the mRNA array database of Oncomine. The results of immunohistochemistry and Western blotting showed that the expressions of Sp1 of colorectal adenocarcinoma tissues and cell lines were highly consistent, but were not different from those of adjacent normal tissues. There was no significant correlation between the expression level of Sp1 protein of colorectal adenocarcinoma tissues and lymph node metastasis, as well as distant metastasis.

Conclusion: The expression of Sp1 of colorectal adenocarcinoma tissues is not significantly different from that of normal tissues and is not significantly correlated to the cancer metastasis.
