Abstract

Therapeutic natural products must eventually be translated to practice. Doing so, however, poses several engineering challenges and opportunities. Most native producers of natural products display characteristics that include slow growth kinetics, immature genetic manipulation protocols, and fastidious nutrient requirements. Such challenges have complicated overproduction process development and spurred new research initiatives such as heterologous biosynthesis. In this chapter, a summary and review of therapeutic natural product production options will be presented. Engineering strategies will be featured at the genetic, metabolic, and process scales. An emphasis will be placed on the influence the producing cellular host, whether native or heterologous, has upon the resulting production process.
