Abstract
A promising therapeutic advance in drug therapy is the prediction of maintenance doses to achieve a targeted steady-state concentration based on a single serum concentration obtained after a test dose. The ability to make predictions in this manner has been based on a linear relationship. Theoretical considerations differ from experimental data and reveal that excessive accumulation of drug may occur outside of a limited and predetermined range of serum concentrations.
