New species of tyrosine tRNA in nonsense suppressor strains of yeast

Abstract

A number of mutations in Saccharomyces cerevisiae are subject to suppression in so-called supersuppressor strains. The tyrosyl-tRNAs from two Class I, Set I ochre-specific suppressor strains, in which UAA is read as tyrosine, have been examined by reversed-phase column chromatography. In each ochre-specific strain there is an extra early-chromatographing tyrosyl-tRNA species present that is not present in wild type. Strains carrying an amber suppressor gene that is allelic to one of these ochre-specific suppressors possess a new late-chromatographing tyrosyl-tRNA not present in the wild type. In addition, strains with either the amber suppressor or its allelic ochre-specific suppressor lack a late-chromatographing portion of the wild-type tyrosyl-tRNA. This may indicate that the new tyrosyl-tRNAs are derived from but one of several presumed components of the main peak of this tRNA.
