Abstract
Multiharmonic Fourier-based phase-and-modulation fluorescence spectroscopy with 257.3 nm excitation is examined. The system exhibits excellent accuracy and picosecond measurement precision: it is capable of rapidly recovering complex fluorescence decays of anisotropy and intensity. The system is well suited for the study of complex fluorophore mixtures, polymer photophysics, and protein photophysics. Photobleaching is not evident in the system, but thermal lensing is common in certain solvent systems when used at 257.3 nm.
