Enzymatic synthesis of 2′-deoxypseudouridylic acid

Abstract

A partially purified pseudouridylate synthetase preparation of Agrobacterium tumefaciens B6 was found to catalyze the reaction between uracil and 2-deoxyribose-5-phosphate. The product of this enzymic reaction was susceptible to cleavage by snake venom 5′-nucleotidase to yield a hitherto undescribed nucleoside, which resembled 8-pseudouridine in its ultraviolet spectral characteristics and its reactivity toward acrylonitrile. The initial synthetic product of the enzymic reaction was identified as 2′-deoxypseudouridylic acid.
