Abstract
The important clinical problems, i. e. the influence of the division of the daily dose and pregnancy on ototoxicity, were studied experimentally. In investigations performed simultaneously, four experimental groups of 10 guinea pigs each received either a single subcutaneous injection of 80 mg tobramycin per kg body weight per day or two subcutaneous injections of the same dose at an interval of 12 hours. The drug was administered for 29 days. A fifth group which was not treated with tobramycin served as the control group. In functional and histological investigations we studied the extent to which dividing the daily dose affected the oto- and nephrotoxicity. In further studies, which were also performed simultaneously, two groups of seven pregnant and non-pregnant guinea pigs each were examined with regard to the influence pregnancy had on the oto- and nephrotoxicity of tobramycin. A decrease in ototoxicity of aminoglycoside antibiotics, which is believed to result from dividing the daily dose, could not be observed. Pregnancy also had no influence on the ototoxicity and nephrotoxicity of tobramycin.
