Abstract

Anti-drug antibodies are increasingly being used within an inverse targeting strategy to increase the pharmacokinetic and therapeutic selectivity of intraperitoneal (i.p.) chemotherapy. The anti-topotecan (TPT) antibodies have demonstrated high affinity and sufficient purity for use in evaluating their use in “inverse targeting” strategies to optimize TPT i.p. chemotherapy. This article describes the development of hybridomas secreting monoclonal antibodies against TPT, and the production, purification, characterization, and pharmacokinetics of an anti-TPT monoclonal antibody and of monoclonal anti-TPT Fab fragments.
