Abstract
The first edition of Comprehensive Medicinal Chemistry was published in 1990 and was intended to present an integrated and comprehensive overview of the then rapidly developing science of medicinal chemistry from its origins in organic chemistry. In the last two decades, the field has grown to embrace not only all the sophisticated synthetic and technological advances in organic chemistry but also major advances in the biological sciences. The mapping of the human genome has resulted in the provision of a multitude of new biological targets for the medicinal chemist with the prospect of more rational drug design (CADD). In addition, the development of sophisticated in silico technologies for structure-property relationships (ADMET) enables a much better understanding of the fate of potential new drugs in the body with the subsequent development of better new medicines. It was our ambitious aim for this second edition, published 16 years after the first edition, to provide both scientists and research managers in all relevant fields with a comprehensive treatise covering all aspects of current medicinal chemistry, a science that has been transformed in the twenty-first century. The second edition is a complete reference source, published in eight volumes, encompassing all aspects of modern drug discovery from its mechanistic basis, through the underlying general principles and exemplified with comprehensive therapeutic applications. The broad scope and coverage of Comprehensive Medicinal Chemistry II would not have been possible without our panel of authoritative Volume Editors whose international recognition in their respective fields has been of paramount importance in the enlistment of the world-class scientists who have provided their individual 'state of the science' contributions. Their collective contributions have been invaluable.
